== Management == Timely management of skeletal dysplasia is important to combat functional deterioration. Due to rarity of the individual disorders that cause skeletal dysplasia, management can be challenging if a patient does not have access to a facility that has physicians who specialize in skeletal dysplasia. Guidelines have been developed for the management different aspects of skeletal dysplasia, including best practices for managing craniofacial and spinal manifestations, diagnosis and management of type II collagen disorders, pregnancy of people with skeletal dysplasia, peri-operative management, and foramen magnum stenosis in achondroplasia.
Despite its sedating action, mirtazapine is also believed to be capable of this, so in the United States and certain other countries, it carries a black box label warning of these potential effects, especially for people under the age of 25. Mirtazapine may induce arthralgia (non-inflammatory joint pain). A case report published in 2000 noted an instance in which mirtazapine counteracted the action of clonidine, causing a dangerous rise in blood pressure. In a study comparing 32 antidepressants of all pharmacological classes, mirtazapine was one of the antidepressants most likely to cause nightmare disorder, sleepwalking, restless legs syndrome, night terrors and sleep paralysis. Surgeons who perform serial sectioning through the block of tissue (usually 100 micrometres apart) are assured of the contiguous nature of his tumor and the distance of the tumor from the surgical margin, and are familiarized with the nature of the tumor. Serial sectioning also makes it easier to work with three-dimensional tumor with margins that are difficult to compress. Compression artifact, freezing artifact, cautery artifact, tissue folds, crush artifact from forceps, relaxing incision artifact, cartilage dropping out, fat compression, poor staining, dropping of tumor, etc. These can be introduced as the tumor is "flattened". Stain can run from the surgical edge, and stain the surgical margin – giving a false impression that the entire surgical margin is clear, when it is not. While some surgeons unfamiliar with the "whole piece" or "PacMan" methods of processing might suggest that multiple piece sectioning is better than one, in fact the more tissue sections are cut, the more artifacts in staining and tissue malformation will be introduced. It is imperative that the surgeon be fully familiar with tissue handling and processing; and not simply rely on a trained technologist to perform their sectioning. The GABA transporter 1 (GAT-1) and GABA reuptake inhibitor tiagabine (Gabitril) is approved and clinically used as an anticonvulsant. It has also been used off-label in the treatment of anxiety disorders and other conditions. The drug increases γ-aminobutyric acid (GABA) levels in the brain and has been found to improve sleep, including by increasing slow wave sleep (deep sleep). In addition, tiagabine has been reported to make sleep feel more restorative and to improve some cognitive outcomes. The drug has an elimination half-life of 5 to 8 hours. While tiagabine may have hypnotic effects, off-label use is discouraged as it has been associated with new-onset seizures in people without epilepsy.

Samidorphan (INNTooltip International Nonproprietary Name, USANTooltip United States Adopted Name) is an opioid antagonist that in the form of olanzapine/samidorphan (sold as Lybalvi) is used in the treatment of schizophrenia and bipolar disorder. Samidorphan reduces the weight gain associated with olanzapine. Samidorphan is taken by mouth. Samidorphan was under development as a standalone medication for various indications but has been discontinued. Buprenorphine/samidorphan for the treatment of major depressive disorder was rejected by the US Food and Drug Administration (FDA) due to insufficient evidence of effectiveness, but remains in preregistration as of September 2021.
