=== Pharmacodynamics === Luvesilocin is a prodrug that is metabolized into 4-HO-DiPT. This metabolite is an analogue of the neurotransmitter serotonin and acts as a non-selective serotonin receptor agonist, including of the serotonin 5-HT2A receptor. Activation of the serotonin 5-HT2A receptor is thought to be specifically responsible for the hallucinogenic effects of serotonergic psychedelics. 4-HO-DiPT produces the head-twitch response, a behavioral proxy of psychedelic effects, in rodents. In drug discrimination tests, 4-HO-DiPT fully substituted for the psychedelic drug DOM, with 5-fold lower potency than DOM and 2-fold lower potency than psilocin (4-HO-DMT). The drug activates basolateral amygdala (BLA) interneurons via the serotonin 5-HT2A receptor to enhance GABAergic inhibition of principal neurons in the BLA, which may mediate an anxiolytic effect of suppression of learned fear (fear extinction) in rodents.
=== Iron chelation === Multiple blood transfusions lead to severe iron overload, as the body eventually breaks down the hemoglobin in donated cells. This releases iron which it is unable to excrete. Iron overload may be treated by chelation therapy with the medications deferoxamine, deferiprone, or deferasirox. Deferoxamine is only effective as a daily injection, complicating its long-term use. Adverse effects include primary skin reactions around the injection site and hearing loss. Deferasirox and deferiprone are both oral medications, whose common side effects include nausea, vomiting and diarrhea.
== Causes == AMP deaminase is an enzyme that converts adenosine monophosphate (AMP) to inosine monophosphate (IMP), freeing an ammonia molecule in the process. It is a part of the metabolic process that converts sugar, fat, and protein into cellular energy. In order to use energy, a cell converts one of the above fuels into adenosine triphosphate (ATP) via the mitochondria. Cellular processes, especially muscles, then convert the ATP into adenosine diphosphate (ADP), freeing the energy to do work. During heavy or prolonged mild to moderate activity, other enzymes convert two molecules of ADP into one ATP molecule and one AMP molecule, making more ATP available to supply energy. AMP is normally converted into IMP by myoadenylate deaminase—so myoadenylate deaminase deficiency reduces energy that would be available to the cell through the purine nucleotide cycle. Instead of being converted to IMP, the AMP builds up in the cells of affected individuals, spills into the blood, and is eventually metabolized in the liver. In persons with a defective enzyme, 5'-nucleotidase removes the ribose and phosphorus from AMP, increasing levels of adenosine measured in muscle cells by ~16–25×, after exercise.
The Product Liability Directive 1985 was the first consumer protection measure. It creates strict enterprise liability for all producers and retailers for any harm to consumers from products, as a way to promote basic standards of health and safety. Any producer, or supplier if the ultimate producer is insolvent, of a product is strictly liable to compensate a consumer for any damage caused by a defective product. In the year 2000, 23 years after production, the radioactive material inside the RTG had decreased in power by 16.6%, i.e. providing 83.4% of its initial output; starting with a capacity of 470 W, after this length of time it would have a capacity of only 392 W. A related loss of power in the Voyager RTGs is the degrading properties of the bi-metallic thermocouples used to convert thermal energy into electrical energy; the RTGs were working at about 67% of their total original capacity instead of the expected 83.4%. By the beginning of 2001, the power generated by the Voyager RTGs had dropped to 315 W for Voyager 1 and to 319 W for Voyager 2. By 2022, these numbers had dropped to around 220 W. NASA has developed a multi-mission radioisotope thermoelectric generator (MMRTG) in which the thermocouples would be made of skutterudite, a cobalt arsenide (CoAs3), which can function with a smaller temperature difference than the current tellurium-based designs. This would mean that an otherwise similar RTG would generate 25% more power at the beginning of a mission and at least 50% more after seventeen years.
